Colony morphology, capsules and negative staining with Indian ink.


Bergey's Manual of Determinative Bacteriology describes colony morphology. Colony morphology is described in terms of form, elevation, margin, surface, opacity and chromogenesis.

The capsule of bacterial colonies, when present, is impermeable to biological dyes and cannot be stained, so negative staining with Indian ink is used. The capsule is then viewed as a clear halo surrounding the bacteria.
